چکیده

Mitochondrial fractions isolated from thoracic muscles of the tobacco horn worm larvae and moths, free of bacterial contaminations, incorporate amino acids into proteins. Optimum activity requires the presence of ATP, an ATPgenerating system, and an amino acid mixture. The pH optimum is around 7.2. The system is not affected by cycloheximide, ribonuclease, or actinomycin D. Puromycin completely inhibits the incorporation, while D-chloramphenico1 reduces the activity by over 50%. The incorporation activity is under physiological control. Mitochondrial fractions from pupae of intermediate developmental stages show the highest activity, while the fractions from the young pupae and from the adult moths exhibit much lower activities.

Further study of factors affecting amino acid incorporation into protein by isolated mitochondria.

1. The incorporation of amino acids into protein in isolated mitochondria has now been studied in more detail, and with mitochondria from a range of tissues. 2. Liver mitochondria from newborn rats are twice as active as those from adult rats. 3. The mitochondria are inactivated by excessive homogenization and repeated freezing and thawing. 4. The incorporation is sensitive to conditions of inc...

Bacterial contamination and amino acid incorporation by isolated mitochondria.

The data presented in this paper emphasize the importance of eliminating bacterial contamination in studies of amino acid incorporation into isolated mitochondria. A correlation between the number of bacteria present and amino acid incorporation into mitochondria was observed when pure cultures of bacteria were added to mitochondria prepared under sterile conditions. Low concentrations of dinit...

Initial stages in the incorporation of amino acids into protein in rat-liver mitochondria.

The addition of cell sap is required for the incorporation of amino acids into rat-liver microsomes and ribosomes (Hoagland, Keller & Zamecnik, 1956; Komer, 1961). The function of the sap involves activation of the amino acids to form amino acyl adenylates (Hoagland et al. 1956) and the subsequent transfer of the activated amino acids to a soluble RNA (Hoagland, Stephenson, Scott, Hecht & Zamec...

Propofol differentially inhibits the release of glutamate, γ-aminobutyric acid and glycine in the spinal dorsal horn of rats

Objective(s): Propofol (2, 6-diisopropylphenol) is an intravenous anesthetic that is commonly used for the general anesthesia. It is well known that the spinal cord is one of the working targets of general anesthesia including propofol. However, there is a lack of investigation of the effects of propofol on spinal dorsal horn which is important for the sensory transmission of nociceptive signal...

The Stimulation by Treatment in Vivo with Tri-iodothyronine of Amino Acid Incorporation into Protein by Isolated Rat-liver Mitochondria.

1. Normal and thyroidectomized rats were treated with near-physiological doses of tri-iodothyronine. Liver mitochondria were isolated and incubated with radioactive amino acids. In normal rats tri-iodothyronine caused only a slight stimulation of incorporation into mitochondrial protein, but in thyroidectomized animals the incorporation was doubled. 2. There was a lag period of about 36 hr. aft...
